Rough Night centers on five female friends and a bachelorette party that goes wrong. The film opens by showing the origins of their friendship when they meet during their freshman year of college at George Washington University. The group eventually grows apart after college, but nearly a decade later, they plan a reunion to celebrate Jess' upcoming wedding. Jess, played by Scarlett Johansson, is a senatorial candidate who is struggling to seem relatable to voters. The five friends convene in Miami for a weekend of partying. Frankie, played by Ilana Glazer, decides to hire a male stripper and from this point, the night goes horribly awry. The four friends realize they must work together to get themselves out of a series of sticky situations.

Lucia Aniello, Paul W. Downs, and Ilana Glazer all got their major starts in comedy together. The three met at the sketch comedy group Upright Citizens Brigade in New York City. When Glazer's comedy show Broad City was picked up by Comedy Central, she recruited friends and frequent collaborators Aniello and Downs to work on the show. Aniello serves as a writer, director and producer for Broad City while Downs acts in addition to writing and producing the show. Glazer herself plays one of the main roles along with writing and executive producing the series.
